You become a vampire by getting infected by fighting a vampire. There's no sure fire way to get infected, it's all random. Forget about it if you've become immune to diseases during the main quest. Make sure you SAVE before you do become one, just incase.

I became a vampire once, more disadvantages than advantages. I made myself a pair of speed boots (kind of like the Boots of Blinding speed without the blind). I know it's kind of cheating but no fast travel was boring the heck outta me. I wouldn't become one again, there's no "great treasure" to be found... although talking to Molag Bal was kind of cool.

You can easily get infected if you have 100% disease resistance, especially if you cast a couple of 100% weakness to common disease spells on yourself.
