I think the best way to make her unique would be to give her a personality/background that offers something different to the mods and vanilla companions already out there. What this could entail is entirely up to you. Good luck, I'm a huge fan of well written follower mods and I'd love to see another one!

Most voiced followers tend to get a reasonably good amount of endorsements depending on the quality of the acting and writing, and having a quest already puts you ahead of several others. Since you're not a scripter you want to have a stand out personality for your follower or some kind of interesting back story as said above and looks will also matter a lot more since people won't be downloading for the advanced scripted features that are unique to certain followers. I'm making my follower unique by having her based around a particular style of humour with many witty remarks and making her a bit socially awkward and inappropriate around others so people will download her (hopefully) because she is very fun to be around and a bit unpredictable and maybe even because they like my sense of humour. So you need a good idea basically and it has to be well written and well executed.

Personally I'd like more unique male follower mods. But if you want to make someone unique in a female follower, then don't make her eye candy like so many others do. Make her a less commonly used race in follower mods, such as Orc, Dunmer or Argonian.

Learn how to do voiceovers or get someone who already does. Don't have to create eyecandy amazon, but don't make them ugly either.

A bit of a throw-back to Baldur's Gate 2, but I love party banter. Emma's Vilja is a perfect translation of this into TES and, IMO, is the best follower mod in both Oblivion and Skyrim. It makes for a far more entertaining character. IMO, any follower lacking this kind of interaction is a waste of time, as the game already includes plenty of boring, non-interactive followers.
I am also keen on keeping to the Lore. Cerwiden is a well-designed follower, but the law-unfriendliness made me hate her. Tying the character to the world and its existing lore is more immersive, and probably a lot easier to do anyway.

Give her a backstory, quests to do, voiced dialog, the works! Make her a bit like the Willow follower mod for New Vegas. Make her interesting, make her someone you can actually care about.
