Just make sure your data files are all loaded, the ones you want anyway, and you can go ahead and play. I don't think you need to worry about your saved game unless the mod says so. I'm just weird so here is a multi-answer: You want to check everytime to make sure your mods are checked so they will run when you start up. If you just added mods and are loading a clean save game it should be safe, wait about 5 game days and some of your mods will refresh inventory lists and the like, while some will be instantaneous.
