Ok, I've gotten all the files and ran everything, what do I do now. How can I get mods like skyUI into the game

I've modded fallout new Vegas but I'm very new to modding Skyrim so I'm really sorry if there is a forum post for this or if its super obvious. But I've been using mod manager, and I downloaded skyUI and Unofficial Skyrim patch. I also installed the Skyrim Script Extender, now I launch using the launcher that comes with the Skyrim Script Extender. But non of my mods do anything, I've also checked the box to enable them. I've seen various posts mentioning making the plugin file in the Skyrim folder not read only, I can't find that file so I wasn't able to do that. Are there any other common reasons that the mods would not do anything? Should I launch in a different way? I've attached a screenshot of my mod organizer, thank you for any and all help I receive!
