First play through the game without mods, to experience the original vision of the developers.
Pushkatu
You should install mods right from the start to save some time, vanilla Oblivion ain't that great anyway, at least install some that will make your game look better, mods like: Natural enviromentsQuarl texture pack - if your PC can handle itone body mod You can still play vanilla, but it will look a lot better. RiskbreakerEnder
I've been on the fence on getting Oblivion lately since I've shredded through Fallout 3 twice. The vanilla run was alright but on my mod run I realized so much more of the game world while slowly modding it up. I'm hoping I can go through the same process in Oblivion if I decide to retry it (tried it briefly on 360 once, it didn't really impress me at that time.) I hope retrying now it'll be like my Fallout experience which took me a long time to get into, but once I finally got to that enlightenment point... That said if I do, as calliton and Monolithic0117 put it, vanilla run first. Mods will be my rewards in the subsequent playthroughs, just to keep things interesting and long lasting.

jaysus
ya gratz on your great taste of booby :P and just use mods right away... oblivion is a darn long game unlike fallout you can really dwell 100h+ in there and you should make that as pleasant as possible and mods are there to do that for you... Certainly you don't mean sinking in 100 hours in the main quest/game path? I was thinking to mod run after MQ.

You don't have to do it to get sucked right into this game, and real-life time sometimes seems to pass as fast as Oblivion time
