If your mod requires OBSE than you must use it every time you use the mod.

I don’t think it matters if you use OBSE & don’t need it.

I think it might depend on the OBSE mod. Since I installed Deadly Reflexes I sometimes get crashes to desktop if I don't go through the loader.

I use a mod that allows me to push people back. It requires OBSE. Sometimes I forget to run OBSE, and the mod just doesn't work. There doesn't appear to be any other side effects, however, this will vary from mod to mod.
