How are you starting it? (from steam or NMM start button)

Are you using skse? If so, are you using the launch skse button?

Are you running both steam and NMM as an administrator?

Did you change any of the file paths for steam or NMM from the default?

Are you logged into steam? If you are using the offline mode, you might need to go back to online for the first time.

I have the same issue after returning home and not playing Skyrim for two weeks. It was running perfectly before with no CTDs. There was a steam client update though before I tried running Skyrim. I tried both with and without the SKSE loaded, ENB injector running, I validated the local files for Skyrim, and even changed the Steam language to German all to no avail. So, if you find the problem and fix it please post it here.

Have you tried renaming (deleting) your My Games\Skyrim\SkyrimPrefs.ini and skyrim.ini to see if the default settings work?

I'm using NMM so I'll probably deactivate all Mods and completely wipe both Skyrim and Steam to force a full download and a full reinstall if all else fails. Then I'll probably find out it was a sound driver issue. :-p

It turns out a mod I updated now requires Dawnguard, the author didn't say so, and didn't even leave the non-Dawnguard version available. So, not having Dawnguard and loading a mod requiring Dawnguard will cause this to happen.

Here is a link to my troubleshooting blog.

This is updated often so come back later for more.

If you use any of this and have some success - or even if nothing works - please post back here what you did that worked or didn't work. That kind of information can be invaluable to other members having the same or similar problems - And to me in fixing any problems in the blog.

That sounds like a potential problem for a lot of people...did you notify the mod author so they can fix their description?
