I've had the infinate loading screen problem hardcore whenever I go to the city. I also have the mysterious serum in my inventory and it doesn't affect my game whatsoever. I think it has something to do with bugged quests. I created a file where you can use a bat command in the console to force complete all minutemen radiant quests. A curious thing about when I used the force complete command I got experience for some unknown reason. I think that there are certain "invisible" quests that float around in the void until you force them to complete. The only thing I've noticed since using my mod is that I don't get radiant minute men quests anymore. A good thing if you ask me. http://www.nexusmods.com/fallout4/mods/10061/? After I used this command I noticed my game doesn't have the infinite loading screen as much. Today I haven't had any at all, which blows my mind. I also think that in the city it may do the infinite loading screen because there is just so much stuff to pack into one place and if your computer isn't super good it will fail on you. I haven't tried dumping some save files so maybe Ill try that next. Would have been nice is Bethesda addressed this glaring loading screen issue. The only thing I can say for certain is that the longer you play the game, the more unstable the game becomes. Small bugs you don't notice build up until it ruins your game. You just have to get rid of as many bugs as you can. Side note: I'm noticing more often than not I'm crashing on buildings directly related to the main quest. Could be a coincidence but who knows.My Institutionalized quest was bugged and I couldn't complete it and I was having this issue. Actually, it's because the RAM for some reason doesn't get flushed and the game waits for the RAM to get flushed so it can load the textures. Usually, I refrain from using the term "game engine bug" but this really is one. Huge Save files, lot of Save files in the "Saves" folder can even catalyze this problem.. But it can happen anytime. I may be wrong but I think there no "deathless" save game. Eventually, all of them will grow too big for the game engine to handle. Of course, DLCs and certain mods will fill up saves more quickly.
