nexus mod manager; launching skyrim

When I click on 'launch skyrim' or 'launch custom skyrim' this error pops up. I assume it's because of my file directory and because I run skyrim off iso mount.

Can anyone please help me solve the problem?

Yes, you need a Steam copy of Skyrim to use the Manager.
